This application was submitted for consideration as a Specialized Center for Research in Pediatric Developmental Pharmacology (RPDP). The goal of the GOLDILOKs program is to investigate the role of ontogeny and genetic variation at the level of drug response. It is based on the concept of response –exposure-dose wherein the desired response is established, the exposure necessary to achieve that exposure for a given patient is determined, and then the dose to achieve that exposure for the individual patient is identified. Essential to this process is the development of drug dosing algorithms to standardize drug exposure such that factor contributing to variability in drug response at the level of the drug target can be identified.
