The proposed research has 2 specific aims: 1) To determine the in vitro and in vivo functions of CRK and CRKL in glioblastoma cell motility using the gene knockdown approach, and 2) To determine the effects of the CRK/CRKL-antagonist peptides on glioblastoma cell motility in cell culture and glioblastoma xenograft mouse models. Successful completion of the proposed research will develop antagonist peptides that inhibit or degrade CRK/CRKL proteins and block glioblastoma cell migration and invasion in vitro and in vivo. The multidisciplinary collaborations for the structure-function relationship studies to develop antagonist peptides and for efficient delivery of the designer peptides into tumor cells and tissues will provide a unique advantage in the development of peptide drugs for glioblastoma.
